Should angiotensin II receptor blockers and statins be combined?

Coronary heart disease (CHD) resulting from atherosclerosis is the single largest killer in the Western world. Approximately 40% of patients with hypertension have hypercholesterolemia, which is central to the pathogenesis of atherosclerosis and cardiovascular disease (CVD).1 Conversely, hypertension is a significant risk factor in patients with elevated cholesterol.2 There is strong synergy be...

Angiotensin II analogs comprised of Pro-Amb (γ-turn scaffold) as angiotensin II type 2 (AT(2)) receptor agonists.

We describe herein the design, synthesis and conformational investigation of Pro-Amb (proline-3-amino-2-methoxybenzoic acid) incorporated Angiotensin II and its truncated analogues. Solution-state NMR and CD studies suggest γ-turn-like conformation in Pro-Amb analogs in aqueous solution. Furthermore, Pro-Amb analogs have been shown to act as AT2 receptor agonists.

Angiotensin II type 2 receptor-bradykinin B2 receptor functional heterodimerization.

Angiotensin II type 2 (AT2R) or bradykinin B2 (B2R) receptor activation enhances NO production. Recently, we demonstrated enhancement of NO production when AT2R and B2R are simultaneously activated in vivo. However, the mechanism involved in this enhancement is unknown. Using confocal fluorescence resonance energy transfer microscopy, we report the distance between the AT2R and B2R in PC12W cel...

Effect of angiotensin II type 1 receptor blockade on cardiac remodeling in angiotensin II type 2 receptor null mice.

To clarify the possible involvement of uninhibited angiotensin II (Ang II) type 2 (AT(2)) receptor stimulation in the effects of an Ang II type 1 (AT(1)) receptor blocker, valsartan, we examined the cardiovascular remodeling induced by aortic banding with the use of wild-type (Agtr2+) and AT(2) receptor null (Agtr2-) mice. Aortic banding caused cardiac hypertrophy in Agtr2+ and Agtr2- mice to a...
